If you are referring to a layer name such as "one two" the answer is
there is no problem. However if you are asking if you can have a total
blank layer name, this can't be done as layer name must have at least
    one character.

  3. Hi,

While AutoCAD itself has no problems, many programs written in the days
before AutoCAD could do this rely on layer names without spaces.

Depending on what customisation you using, I would not use layer names
    without spaces by choice.

It all depends on the AutoCAD version, and possibly the value of EXTNAMES.

Use the (snvalid) function to tell you if a symbol name is valid or not.
Note that the sysvar EXTNAMES is used to determine which set of rules
    that (snvalid) will follow.
